Abstract

Introduction: Neuroinflammation has emerged as a crucial factor in understanding depression, a condition that affects approximately 264 million people worldwide, according to data from the World Health Organization. The relationship between inflammation and mental health is a growing field of research that seeks to understand how inflammatory processes can influence the severity and treatment response of depression. Objective: This study aims to analyze the intersection between neuroinflammation and depression, emphasizing the importance of inflammatory biomarkers in the pathogenesis of depression and the enhancement of therapeutic interventions. Methodology: A narrative review was conducted using the PubMed platform, employing the search strategy "Inflammation OR neuroinflammation AND depression OR mood disorders AND anti-inflammatory agents." Filters were applied to restrict the search to clinical trials, randomized clinical trials, reviews, and systematic reviews with meta-analysis published between 2019 and 2025. Following an initial screening of 142 studies, 19 articles were selected for detailed analysis. Results: The activation of glial cells, such as microglia and astrocytes, and the release of inflammatory cytokines, such as interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α), play significant roles in the severity of depressive symptoms. Elevated levels of inflammatory biomarkers, such as C-reactive protein (CRP), are associated with greater symptom severity, suggesting that the assessment of these markers may be useful in patient stratification and treatment personalization. Conclusion: The identification of inflammatory biomarkers holds promise for enhancing the diagnosis and management of depression, indicating that anti-inflammatory interventions may improve clinical outcomes. Understanding the role of neuroinflammation in depression not only enriches knowledge about its pathophysiology but also opens new avenues for the development of targeted therapies, thereby contributing to global mental health.
