Abstract

This study examines the pathological mechanisms and potential therapies for Chronic Traumatic Encephalopathy (CTE), a neurodegenerative disease associated with repetitive head trauma, commonly seen in contact sports and high-risk professions. The research focuses on understanding the aggregation of hyperphosphorylated tau protein and chronic brain inflammation, both of which play a central role in disease progression. The objective is to contribute to the field of CTE by deepening the analysis of the underlying pathological mechanisms and exploring potential therapeutic interventions. An integrative literature review was conducted, encompassing studies published between 2012 and 2024 in Portuguese, English, and Spanish. This synthesis provides insight into CTE development and promising therapeutic interventions. Results suggest that therapies targeting tau modulation and anti-inflammatory interventions show potential in preclinical studies but require further robust clinical validation. Diagnostic techniques, such as biomarkers and positron emission tomography (PET), are emerging as promising tools for early detection but also require standardization through additional research. The review highlights the need for further studies to confirm the efficacy of these interventions and improve the clinical management of CTE. It concludes that continued research is crucial to developing more effective treatments and mitigating the impact of this condition on public health, particularly in young and high-risk populations.
